猪圆环病毒2型青海分离株的分子鉴定及进化分析

      本研究对青藏高原地区猪圆环病毒青海分离株(PCV2-QH1)进行了Cap基因的克隆鉴定及测序分析,并与我国及其他国家或地区的毒株进行了同源性比较和遗传进化分析。结果显示:PCV2-Cap基因核苷酸序列全长705 bp,编码蛋白为234个氨基酸,分子量大小约28.06 kDa,为不稳定蛋白,总体亲水性非常高,可溶于水;PCV2-Cap蛋白二级结构以无规则卷曲为主,有多个有效的抗原表位;PCV2-QH1分离株与其他参考序列的核苷酸和氨基酸序列同源性均在99%以上;PCV2-QH1分离株与PCV2美国分离株(KU697156)组成一个微小分支,并与美国毒株、韩国毒株聚在一支,表明它们之间的亲缘关系最近。本次对青海分离株PCV2-QH1的鉴定,可为我国猪圆环病毒分子流行病学调查提供数据参考。
英文摘要:
      In this study,Cap gene of porcine circovirus Qinghai strain(PCV2-QH1)was cloned and sequenced,meanwhile,the homology comparison and evolution analysis were carried out between PCV2-QH1 and strains from other regions or countries. Results showed that the Cap gene was 705 bp in length,encoding 234 amino acids,and the molecular weight of Cap protein was 28.06 kDa. The protein was not stable with high hydrophilicity and was soluble in water. The main secondary structure of PCV2-Cap protein was irregular coiled-coil,with multiple effective epitopes. The sequence homology of the PCV2-QH1 with other referenced strains were all above 99%. Besides,the PCV-QH1 isolate and the PCV2-USA isolate(KU697156)formed a tiny branch,and PCV-QH1 isolate was clustered together with the USA and South Korea strains,indicating a close relationship. As a conclusion,the study would provide data reference for epidemiological investigation of PCV in China.
