    为提高鸡血浆中禽白血病病毒(avian leukosis virus,ALV)的检出率,提高净化效果,本试验随机选择94只225日龄淘汰公鸡,采集血浆分别采用常规的离心法和改进的自然沉淀法进行血清分离、DF-1细胞培养,然后采用ELISA试剂盒检测ALV P27抗原,并对检测结果进行SPSS对比分析后利用改进法对两个品种鸡群开展净化检测。结果显示:改进法(A组)细胞培养状态优于常规法(B组),A组P27抗原检测阳性率比B组提高了6.39%,两组的OD值差异不显著(P>0.05),净化检测可基本实现ALV净化。结果表明,改进法省去了离心流程,有利于DF-1细胞的生长和维持,并提高了检出率,从而可以加快ALV净化进程。

    Abstract:

    In order to improve the detection rate of avian leukosis virus(ALV)in chicken plasma and increase the effectiveness of purification,94 eliminated cocks at 225-day old were randomly selected to collect plasma,followed by serum isolation by conventional centrifugation and optimized natural precipitation method,and culture of DF-1 cells,then ALV P27 antigen was detected by ELISA kits,and the two breeds of cocks were purified and tested by the optimized method after the results were compared and analyzed by SPSS. The results showed that the status of cell culture(Group A)by the optimized method was better than that by the conventional method(Group B),the positive rate of P27 antigen in Group A was 6.39% higher than that in Group B,the difference between the OD values of the two groups was not significant(P>0.05),and ALV could be purified in general. It was concluded that the optimized method without the centrifugation process was conductive to the growth and maintenance of DF-1 cells,and improved detection rate,which could speed up the purification process of ALV.
