球孢白僵菌对小亚璃眼蜱的毒力测定及其孢外水解酶活性分析

    摘要:

    球孢白僵菌作为一种很重要的生防真菌,已被广泛用于农业害虫和蜱的防治。在侵染宿主的同时,球孢白僵菌分泌大量孢外水解酶,如孢外蛋白酶和几丁质酶。这些酶类不但能够降解宿主表皮,而且被认为是菌株致病性的关键决定因子。为获得高毒力菌株的筛选方法,选取实验室分离鉴定的5株球孢白僵菌菌株,通过测定菌株的孢外水解酶活性和菌株对小亚璃眼蜱的致死率,并对球孢白僵菌的毒力和酶活性进行了相关性分析。结果显示,菌株蛋白酶活性与菌株毒力无相关性,菌株几丁质酶活性与菌株毒力呈现负相关性。本研究为蜱生防高毒力菌株的筛选以及酶活性的初步分析提供了理论依据。

    Abstract:

    Beauveria bassiana(B. bassiana),as a very important biocontrol fungi,has been widely used to control pest insects and ticks in agriculture. When it is invading into any host,a large number of extracellular hydrolases were secreted,such as extracellular proteases and chitinases,which not only degrade the epidermis of the host,but also are considered as the key pathogenicity determinants. In order to identify a method to screen the strains with high virulence,five strains of B. bassiana isolated and identified in laboratory were used to determine the activity of extracellular hydrolase and their lethality to Hyalomma anatolicum anatolicum,then the correlation between their virulence and enzyme activity was analyzed,and no correlation was observed,but a negative correlation between the activity of chitinase and the virulence was found. Therefore,a theoretical basis was provided to screen the strains with high virulence and to preliminarily analyze enzyme activity.
