    2020年11月,上海市某宠物医院接诊一例疑似猫慢性牙龈炎(feline chronic gingivostomatitis,FCGS)病例。为确定病因,采集该病例的眼、口、鼻拭子进行病原分离鉴定,结果确定为猫杯状病毒(feline calicivirus,FCV)和细菌的混合感染。对FCV分离株VP1基因进行扩增和测序分析,发现其与其他18株序列的核苷酸同源性为74.2%~81.0%,编码蛋白氨基酸同源性为82.2%~88.1%。构建进化树分析发现,FCV分离株与国内毒株的遗传关系较国外更近,但与目前临床上使用的疫苗株处于不同的进化分支。对VP1 蛋白D区和E区分析发现,D区及conE区的线性表位较为保守,而E区的5'HRV表位存在较大变异,与抗体反应有关的第439~441位氨基酸同样变异较大。本研究为了解FCV的变异特点以及该病的临床防治提供了参考。

    Abstract:

    In November 2020,a case of suspected feline chronic gingivostomatitis(FCGS)was received by a pet hospital in Shanghai. In order to identify possible causes,swabs were collected from the eyes,mouth and nose of the infected animal for pathogen isolation and identification,which was confirmed to be a mixed infection with feline calicivirus(FCV)and bacteria. It was found that,through amplification and sequencing of the VP1 gene of FCV isolate,its nucleotide homology with other 18 strains was 74.2% ~81.0%,and the amino acid homology of coding protein was 82.2% ~ 88.1%. It was analyzed that,through phylogenetic tree,the isolate shared closer genetic relationship with domestic strains compared to foreign strains,but it was in a different evolutionary branch from the vaccine strains currently used in clinical practice. The regions of D and E of VP1 protein were analyzed,and it was found that the linear epitopes in the regions of D and conE were relatively conservative,while 5'HRV epitopes in E region mutated greatly,so did the 439~441th amino acids related to antibody response. A reference was thus provided for understanding the variation characteristics of FCV and for preventing and controlling the disease.
