    摘要:

    鸡传染性支气管炎(avian infectious bronchitis,IB)具有极强的传染性,为我国二类动物疫病。IB的准确诊断对其防控具有极其重要作用。本文综述了病毒分离、血清学诊断、免疫组织化学技术、分子生物学诊断、蛋白芯片技术等主要的IB诊断方法。病毒分离是诊断IB的重要手段,但其耗时长且对操作有一定要求,因此近年来将其与基因测序分析相结合来进行IB诊断及基因型鉴别;在常规血清学方法中,酶联免疫吸附试验(ELISA)和血凝抑制试验(HI)是推荐用于鸡群大规模筛查及免疫效果评价的方法,病毒中和试验(VNT)具有高度特异性,常被应用于毒株的变异鉴定,但是耗时且缺乏标准化,建议选用更成熟的ELISA方法;随着分子生物学技术的发展,聚合酶链式反应(PCR)因具有更高的准确性,且能实现实时定量分析,已被广泛应用于实验室检测;重组聚合酶等温扩增技术(RPA)特异性好,对试验条件要求不高,适用于养殖场的大规模IB筛查。另外,蛋白芯片技术、免疫层析试纸条、鸡声信号分析装置也为IB诊断提供了全新的思路。

    Abstract:

    Avian infectious bronchitis(IB)is a highly contagious disease under class II animal diseases in China. An accurate diagnosis of IB plays an extremely important role in its prevention and control. In the paper,major diagnosis methods such as virus isolation,serological diagnosis,immunohistochemical technology,molecular biological diagnosis and protein microarray were summarized. Virus isolation was an important means to diagnose IB,but it was time-consuming and required certain operation conditions,so it had been used together with gene sequencing analysis for IB diagnosis and genotype identification;for routine serological methods,enzyme linked immunosorbent assay(ELISA)and hemagglutination inhibition test(HI)were recommended for large-scale screening and immune effect evaluation for chickens,virus neutralization test(VNT)was often used for variation identification of virus strains due to its high specificity,but it was time-consuming and lack of standardization,so a more mature ELISA was recommended;with the development of molecular biology technology,PCR technology had been widely used in laboratory detection as it was with higher accuracy for real-time quantitative analysis;and recombinase polymerase amplification(RPA)was appropriate for large-scale IB screening in farms due to its good specificity and low requirements for test conditions. In addition,a new idea for IB diagnosis was also provided by protein microarray,immunochromatographic strip and device for analyzing chicken sound signal.
