摘要:In order to analyze sequence similarities of hemagglutinin(HA) gene among avian influenza virus(AIV) subtype H5N2 isolates, the HA gene of avian influenza virus A/Ostrich/Denmark/72420/96 (LPAI-H5N1) isolate was amplified by RT-PCR according to the sequence published on GenBank.Then, the sequences were compared with GenBank data. The result showed that the cDNA contained 1737bp covering the whole ORF of HA gene. The HA gene sequence of the isolated strain shared about 80% nudleotide sequence identity with H5N1 isolates, but about 97% with some H5N2 isolates.The results confirmed the high frequence of recombination of the eight segment of AIV genome and the high variation ability of AIV. The amino acids of the cleavage site were E----T-R, confirmed that LPAI-H5N1 was a low pathogenic strain. The amino acid sequence shared about 90% identity with H5N1 isolates and the antigenicity was almost the same. So the H5N2 vaccine can protect the chickens from H5 subtype infection.

摘要:We isolated blood samples from 171 sheep representing the eight main local sheep breeds in the Xinjiang Autonomous Region to identify the PRNP polymorphisms and to determine whether these breeds were at risk for developing scrapie. After DNA extracting and gene sequencing, polymorphisms were found at condons 136(A/A),154(H/R)and 171(Q/R/H/K). However, polymorphisms of valine(V) at codon 136 were not observed, showing that the PRNP genotypes in Xinjiang local sheep breeds which we collected were type A scrapie resistant genotypes.

摘要:The gE gene of PRV La strain is amplified and sequenced. The ORF of PRV LA gE gene is 1740bp-coding glycoprotein E. The homology of deduced amino acids of PRV La strain with other PRV strains is high. The variance of PRV gE sequence shows regular regionally. PRV gE genes shared limited homology to other alphaherpesviruses equivalents: HSV-1?BHV-1?EHV-1?CHV-1?SHV-SA8?SVV?MDV-1 and ILTV. The homology analysis of deduced amino acids of above alphaherpesviruses equivalents showed that gE could be separated into 4 groups: simplex?varicella?Marek's disease-like viruses and infectious laryngotracheitis virus group,the phylogenetic tree analysis contribute further evidence of the evolution of gE of alphaherpesviruses from a common ILTV gE progenitor,and evolutional trend of gE conforms with the evolution of their hosts`.
